Design: DNA extracted using 100/G columns (Qiagen), sheared using a Corvaris g-tube, and libraries made using the PacBio SMRTbell Template Prep kit 1.0. The libraries are size selected to 15 kb using the BluePippin and loaded onto a PacBio RS II.

Study: Comparison of related Escherichia coli O157:H7 strains

The aim of this project is to compare complete closed genomes with whole genome sequencing (WGS) using short reads. SNPs from WGS is used in many studies to determine relatedness of strains. These SNPs are in the core genome and don't reflect the diversity of the complete genome. This study provides insight into how core SNPs relates to the structure and sequence of the complete genome.
